Expression of Bacillus thuringiensis insecticidal protein gene in transgenic oil palm Electron. J. Biotechnol.
Lee,Mei-Phing; Yeun,Li-Huey; Abdullah,Ruslan.
Oil palm, like all other crops, is susceptible to attack from several insect pests causing significant reduction in productivity. In the past, cry genes from Bacillus thuringiensis have been reported to be effective in conferring resistance towards insect pests in crops such as corn and rice. One of the advantages of these toxin proteins is their specificity towards certain harmful insects. A rapid and efficient method was developed for the transformation and evaluation of CryIA(b) expression in oil palm. A recombinant vector was introduced into immature embryos (IEs) of oil palm via the biolistic method. More than 700 putative transformed IEs from independent transformation events were generated. Transient transformation efficiency of 81-100 % was...

Genetic parameters and selection strategies for soybean genotypes resistant to the stink bug-complex Genet. Mol. Biol.
Godoi,Cláudio Roberto Cardoso de; Pinheiro,José Baldin.
Soybean genotypes resistant to stink bugs are derived from complex breeding processes obtained through indirect selection. The aim of the present work was to estimate genetic parameters for guiding selection strategies towards resistant genotypes, based on those traits associated with responses to pod-attacking stink bugs, such as the grain filling period (GFP), leaf retention (LR), percentage index of pod damage (PIPD) and percentage of spotted seeds (PSS). We assessed the parental lines IAC-100 (resistant) and FT-Estrela (susceptible), the progenies F2 and F4, 30 progenies F2:3, 30 progenies BC1F2:3 and 30 progenies BC2F2:3, besides the cultivars BRS Celeste and MGBR-46 (Conquista). Three field experiments, using randomized complete block design with...

Hierarchical and optimization methods for the characterization of tomato genotypes AGRIAMBI
Peixoto,Joicy V. M.; Almeida,Rafaela S. de; Rocha,Jaine P. R. da; Maciel,Gabriel M.; Santos,Nádya C.; Pereira,Lucas M..
ABSTRACT The correct characterization of germplasm banks is fundamental for breeders to succeed in breeding programs. Several studies have sought to obtain genotypes with resistance to pests. However, there is no consensus about which methodology is the most appropriate to characterize a germplasm bank of tomato with different levels of resistance to pests. The objective of this study was to compare methods of multivariate analysis for the evaluation of genetic diversity in tomato genotypes with different levels of resistance to pests. The experiments were conducted at the Vegetable Experimental Station of the Federal University of Uberlândia - Monte Carmelo campus (18º 42’ 43.19” South latitude and 47º 29’ 55.8” West longitude, 873 m altitude), in the...

Insect resistance and risk assessment studies of advanced generations of basmati rice expressing two genes of Bacillus thuringiensis Electron. J. Biotechnol.
Rahman,Mahmood-ur; Rashid,Hamza; Shahid,Ahmad Ali; Bashir,Khurram; Husnain,Tayyab; Riazuddin,Sheikh.
Advanced generations of different transgenic lines of indica basmati rice (Basmati-370) expressing two unrelated Bt genes, cry1Ac and cry2A were evaluated for resistance to Yellow Stem Borer (YSB) and Rice Leaf Folder (RLF) under field conditions compared to control lines over three years (2003-2005). Homozygous lines were selected and analyzed for insect resistance, morphological, physiochemical properties and risk assessment studies. After artificial infestation of target insects, the transgenic plants showed significant resistance. Data were recorded in terms of dead hearts and white heads at vegetative and flowering stage respectively. Transgenic lines showed up to 100 and 96% resistance against yellow stem borer at vegetative and flowering stages,...

Resistance to Anticarsia gemmatalis Hübner (Lepidoptera, Noctuidae) in transgenic soybean (Glycine max (L.) Merrill Fabales, Fabaceae) cultivar IAS5 expressing a modified Cry1Ac endotoxin Genet. Mol. Biol.
Homrich,Milena Schenkel; Passaglia,Luciane Maria Pereira; Pereira,Jorge Fernando; Bertagnolli,Paulo Fernando; Pasquali,Giancarlo; Zaidi,Mohsin Abbas; Altosaar,Illimar; Bodanese-Zanettini,Maria Helena.
Somatic embryos of the commercial soybean (Glycine max) cultivar IAS5 were co-transformed using particle bombardment with a synthetic form of the Bacillus thuringiensis delta-endotoxin crystal protein gene cry1Ac, the beta-glucuronidase reporter gene gusA and the hygromycin resistance gene hpt. Hygromycin-resistant tissues were proliferated individually to give rise to nine sets of clones corresponding to independent transformation events. The co-bombardment resulted in a co-transformation efficiency of 44%. Many histodifferentiated embryos and 30 well-developed plants were obtained. Twenty of these plants flowered and fourteen set seeds. The integration and expression of the cry1Ac, gusA and hpt transgenes into the genomes of a sample of transformed...

Revisiting the Impact of Bt Corn Adoption by U.S. Farmers AgEcon
Fernandez-Cornejo, Jorge; Wechsler, Seth James.
This study examines the impact of adopting Bt corn on farm profits, yields, and insecticide use. The study employs an econometric model that corrects for self-selection and simultaneity. The model is estimated using nationwide farm-level survey data for 2005. Regression analysis confirms that Bt adoption is associated with increased profits, yields and seeding rates. However, the results of this analysis suggest that Bt adoption is not significantly related to insecticide use. This result appears to be related to the fact that insect infestation levels were lower in 2005 than they were in earlier years.

The Emergence of Insect Resistance in Bt-Corn: Implication of Resistance Management Information Under Uncertainty AgEcon
Linacre, Nicholas A.; Thompson, Colin J..
The successful management of transgenic technology is likely to depend on the economic behavioral response of farmers to the regulated use of transgenic crops. A well-studied example is the widespread use of Bt-corn, in the United States, and elsewhere, to control the European Corn Borer, a major corn pest. The extensive use of Bt-corn has led to concerns about the emergence of insect resistance. The United States Environment Protection Agency addressed this potential problem by developing an insect resistance management strategy, based, in part, on complex mathematical models using detailed biological assumptions about the population genetics and life history of the European Corn Borer. However, seed companies and others have sometimes used simpler...

Bahng Taen is a non-glutinous, non-photoperiod sensitive, short duration and high yielding rice variety. This variety showed resistance against brown planthopper (BPH) in the Eastern region, but the resistance has decreased drastically. Therefore, marker-assisted backcrossing was applied to improve BPH resistance by introgression of a Bph3 resistant gene from PTB33 and Rathu Heenati into Bahng Taen. Two SSR markers, RM 170 for Bahng Taen with PTB33 crossing and RM 190 for Bahng Taen with Rathu Heenati crossing, were used in combination with plant type selection to obtain the target plants with Bph3 gene. Twenty six and thirty heterozygous BC4F1 plants were developed from the cross of Bahng Taen with PTB33 and Bahng Taen with Rathu Heenati, respectively....
